Abstract

Die vorliegende Arbeit beinhaltet die Totalsynthese der Naturstoffe und 3-Acyltetramsäuren Macrocidin A und F-14329. Beide Verbindungen sind sind Tyrosin-abgeleitete Tetramsäuren. Macrocidin A, ein para-Cyclophan, ist ein 17-gliedriger Macrocyclus, welcher eine selektive herbizide Wirkung gegen einige Unkräuter besitzt. Erstmalig wurde er mit anderen Metaboliten aus dem Pilz Phoma Macrostoma isoliert. Die Retrosynthese spaltet den Ring in zwei Hälften, einen Tetramsäure- und einen Carbonsäureteil. Nach seperater Synthese beider Moleküle, können mittels eines Yoshii-Yoda-Protokolls beide Teile zu einer 3-Acyl-Tetramsäure verknüpft werden und anschließend in einer seltenen Williamson-Makroveretherung der Ring geschlossen werden. Aufgrund der seperaten Synthese von Tetram- und Carbonsäure und der späten Verknüpfungsschritte, weist die Synthese einen modularen Charakter auf. Die Tetramsäure F-14329 gehört zu der Klasse der beta-Hydroxy-3-Acyl-Tetramsäuren, zu welchen bis zu dieser Arbeit noch kein totalsynthetischer Zugang veröffentlicht wurde. Die zusätzliche Hydroxyfunktion erschwert die Synthese im erheblichen Maße. Mittels Evans-Auxiliar- und Schöllkopf-Auxiliar-Methoden konnten die notwendigen Motive des Naturstoffes synthetisiert werden. Nach Verknüpfung zum beta-Ketoamid und Dieckmann-Cyclisierung konnte der gewünschte, geschützte Naturstoff erhalten werden. Aufgrund der Reaktivität des Systems und gezielten Vorexperimenten, musste eine photolabile Schutzgruppe, ortho-Nitrobenzyl, für den Tetramsäurestickstoff verwendet werden, welche in diesem Zusammenhang erstmalig zum Einsatz kam. Diese notwendige Schutzgruppe kann unter neutralen Bedingungen abgespalten werden. Dieses allgemeine Protokolle ermöglicht die Synthese weiterer Vertreter der Klasse der beta-Hydroxy-3-Acyl-Tetramsäuren.

The present work contains the total synthesis of the natural products and 3-acyltetramic acids macrocidin A and F-14329. Both compounds are tyrosine-derived tetramic acids. Macrocidin A, a para-cyclophane, is a 17-membered macrocycle which has a selective herbicidal effect against some weeds. It was first isolated from the fungus Phoma Macrostoma with other metabolites. The retrosynthesis cleaves the ring into two parts, a tetramic acid part and a carboxylic acid part. After separate synthesis of both molecules, both parts can be linked to a 3-acyl-tetramic acid by means of a Yoshii-Yoda protocol and then the ring is closed in a rare Williamson macroetherification. Due to the separate synthesis of tetram- and carboxylic acid and the late macroetherification, the synthesis has a modular character. Tetramic acid F-14329 belongs to the class of beta-hydroxy-3-acyl-tetramic acids to which up to this work no total synthetic approach has yet been published. The additional hydroxy function makes synthesis considerably more difficult. By means of Evans-auxiliary and Schöllkopf-auxiliary methods the necessary motifs of the natural product could be synthesized. After connection to the beta-ketoamide and Dieckmann cyclization, the desired, protected natural product could be obtained. Due to the reactivity of the system and prior experiments, a photolabile protective group, ortho-nitrobenzyl, had to be used for the tetramic acid nitrogen, which was used for the first time in this context. This necessary protecting group can be cleaved under neutral conditions. This general protocol allows the synthesis of further representatives of the class of beta-hydroxy-3-acyl-tetramic acids.
