Abstract

Die Motivation der vorliegenden Arbeit ergab sich aus den Vorarbeiten zur Totalsynthese von (−)-Combretastatin D-1 am Arbeitskreis Schobert. Der Schlüsselschritt der Synthese ist der Ringschluss. Dieser sollte durch eine intramolekulare Addition-Wittig- Reaktion der Cyclisierungsvorstufe mit Ketenylidentriphenylphosphoran zum Makrolacton erfolgen. Es stellte sich heraus, dass die Cyclisierung nicht erreicht werden konnte, stattdessen das entsprechende Dimer gebildet wurde. An dieser Stelle rückte die Synthese von Combretastatin D-4, dem einfachsten Vertreter der D-Combretastatine in den Mittelpunkt dieser Arbeit. Nachdem Cyclisierungsversuche via Wittig-Reaktion oder Metathese gescheitert waren, wurde geplant, einen größeren Ring zu erzeugen, um dann durch Ringkontraktion den gewünschten Makrocyclus zu erhalten. Dazu wurde die Ramberg-Bäcklund-Reaktion (RBR) ausgehend von Sulfon 132 untersucht. Die RBR von wurde unter verschiedenen Reagenzien und Bedingungen, sowohl in der klassischen 2-stufigen Variante als auch in der in situ Variante, ausgeführt. Trotz zahlreicher Versuche gelang die Darstellung der Zielverbindung auf diesem Weg nicht. Daher wurde eine weitere Möglichkeit zur Ringkontraktion untersucht. Die Darstellung von 11-O-Methyl Corniculatolid A wurde erfolgreich durch Sulfonpyrolyse realisiert. Die Sulfonpyrolyse wurden von Dr. Alan Aitken an der Universität von St. Andrews durchgeführt. Ein optimiertes Verfahren zur finalen Spaltung des Methylethers lieferte Combretastatin D-4 in 97 % Ausbeute. Die Gesamtausbeute der Synthese von Combretastatin D-4 betrug damit 20 % über 7 Stufen ausgehend von Diarylether 127. Im Rahmen dieser Dissertation wurde erfolgreich gezeigt, dass die Synthesestrategie via Sulfonpyrolyse auch auf weitere Combretastatine der D-Reihe übertragbar ist. Neben der Totalsynthese für 11-O-Methyl-Corniculatolid A und Combretastatin D-4 gelang mit dieser Strategie eine enantioselektive Formalsynthese für (−)-Combretastatin D-1, sowie die Totalsynthese von Combretastatin D-2. Die Sulfonpyrolyse zum Combretastatin D-2 lieferte neben der Zielverbindung ein weiteres Pyrolyseprodukt, dessen Struktur nach röntgenkristallographischer Analyse zweifelsfrei bestätigt wurde. Für derartige cyclische Diarylether, mit fünf Kohlenstoffen in der Kette, gibt es in der Literatur keine vergleichbare Verbindung. Wohingegen entsprechende Makrocyclen mit sieben Kohlenstoffen in der Kette zu genüge bekannt sind und in der Literatur als Diarylether- Heptanoide (DAEH) bezeichnet werden. The motivation of this thesis arose from preliminary investigations of the total synthesis of (−)-combretastatin D-1 by the Schobert´s group. The cyclisation remains the key step for synthesis of D-1. A Diarylether should be cyclized to the macrolactone by an intramolecular addition-Wittig-cascade with ketenylidentriphenylphosphorane. Unfortunately, numerous attempts only furnished the unwanted dimer and no trace of the desired compound was observed. At this point of the thesis the synthesis of the simplest congener of the combretastatins D, combretastatin D-4, was focused. As neither the intramolecular Wittig-reaction nor a RCM approach was successful, a completely new synthetic strategy was considered. This new strategy relies on obtaining the target compound by ring contraction of a larger cycle. Therefore the Ramberg-Bäcklund reaction (RBR) was performed by sulfone 132. The RBR was tried under various reaction conditions. Neither the classic two-step process nor the in situ variation of this key transformation provided the ring-contracted compound after numerous efforts. So another approach for the ring contraction of the sulfone was necessary. The preparation of 11-O-methyl corniculatolide A was successfully realized by sulfone pyrolysis which was performed by Alan Aitken at the University of St. Andrews. The procedure for the final demethylation step was optimized to reveal combretasatin D-4 in 97% yield. Combretastatin D-4 could be synthesized in a straightforward fashion from diarylether 127 over seven steps in 20% overall yield. In the course of this thesis it has appeared that the sulfone pyrolysis approach has a great capability in the straightforward synthesis of other natural products of the combretastatin D family. In addition to the total synthesis of 11-O-methyl corniculatolide A and combretastatin D-4, an enantioselective formal synthesis for (−)-combretastatin D-1 as well as the total synthesis for combretastatin D-2 was accomplished using this approach. Beside the desired O-methyl-combretastatin D-2 the sulfone pyrolysis also led to the interesting byproduct via further CO2 extrusion. The structure of this novel compound was doubtlessly confirmed by x-ray crystallographic analysis. No comparable cyclic diarylethers of this type consisting of five carbons in the alicyclic chain are known to date, while analogous diarylethers with seven carbons in the alicyclic fragment are well known and referred as diarylheptanoide (DAEH). So the new compound is very interesting as it represents the first diarylpentanoide (DAEP) with an oxa[1.5]metaparacyclophane scaffold.
