Abstract

Die vorliegende Arbeit beschreibt die Totalsynthesen des Sesquiterpenchinons Spongiachinon (1) und der Sesquiterpenhydrochinone (±)-Siphonodictyal B Derivat (±)-55 und (±)-Wiedendiol B ((±)-2). Spongiachinon (1) Schlüsselverbindungen bei der Synthese des Spongiachinons (1) waren die diastereomeren Benzylalkohole 5a,b. Ausgehend von (-)-Albicansäure ((-)-7) wurde über den (-)-Albicansäuremethylester ((-)-6) durch Reduktion mit DIBAL (+)-Albicanol ((+)-11) erhalten. PCC-Oxidation von (+)-11 lieferte (-)-Albicanal ((-)-3), das mit lithiiertem Tetramethoxybenzol zu den diastereomeren Benzylalkoholen 12a,b gekuppelt wurde. Katalytische Hydrierung von 12a,b an Palladium auf Kohlenstoff ergab die gewünschte Zwischenstufe 5a,b. Acetylierung von 5a,b zu 17a,b und anschließende CAN-Oxidation führte zu dem o-Dimethoxybenzochinon 19a,b. Durch Umsetzung des o-Dimethoxychinons 19a,b mit p-Toluolsulfonsäure konnte Spongiachinon (1) erhalten werden. Die Gesamtausbeute der ersten Totalsynthese von Spongiachinon betrug ausgehend von (-)-Albicansäure ((-)-7) 21 % über acht Stufen. Die Umsetzung der diastereomeren Benzylalkohole (±)-5a,b mit Trichlortrifluoraceton in Gegenwart von p-Toluolsulfonsäure führte zum 5,10-Drimenylaren (±)-15, das durch eine interessante Umlagerungsreaktion entstand. CAN-Oxidation von (±)-15 ergab das o-Dimethoxybenzochinon (±)-22, das durch Behandlung mit Perchlorsäure (±)-Isomamanuthachinon ((±)-20) lieferte. Siphonodictyal B Derivat (±)-55 Für die Synthese des Siphonodictyal B Derivates (±)-55 wurde ein neues Kupplungsverfahren eingesetzt. Dazu wurde (±)-Drimansäurechlorid ((±)-28) mit entsprechend geschützten lithiierten Aromaten umgesetzt, die aus den Bromiden 32, 37, 45, 56 und 13 durch Brom-Lithiumaustausch mit n-BuLi gebildet wurden. Die Darstellung der Aromatenbausteine gelang häufig über deutlich effektivere und kürzere Synthesewege im Vergleich zu Literaturverfahren. (±)-Drimansäurechlorid ((±)-28) wurde aus (±)-Drimansäure ((±)-29) durch Reaktion mit Oxalylchlorid / DMF in quantitativer Ausbeute erhalten. Die durch die Kupplung entstandenen Ketone konnten alle mit LiBHEt3 in Ausbeuten von 100 % zu den diastereomeren Benzylalkoholen reduziert werden. Die Einführung der Formylgruppe in Position 3' durch Lithiierung und Umsetzung mit DMF gelang nur bei Styren (±)-53, das aus Keton (±)-51 durch Reduktion zu den Benzylalkoholen (±)-52a,b und anschließende Eliminierungsreaktion gebildet wurde (Abb. 5.4). Demethylierung von Aldehyd (±)-54 mit Bortrichlorid / Tetrabutylammoniumiodid ergab das Siphonodictyal B Derivat (±)-55, das bisher nicht zu Siphonodictyal B entschützt werden konnte. (±)-Wiedendiol B ((±)-2) Zur Darstellung von (±)-Wiedendiol B ((±)-2) wurde der Aromat 57 mit t-BuLi lithiiert und anschließend mit (±)-Drimansäurechlorid ((±)-28) zum Keton (±)-63 gekuppelt. Reduktion der Ketofunktion mit Lithiumtriethylborhydrid lieferte die diastereomeren Benzylalkohole (±)-64a,b. Für die Eliminierung von Wasser aus den Benzylalkoholen (±)-64a,b wurden zwei neue Verfahren entwickelt, die mit Chloral / p-Toluolsulfonsäure und dem Pyridin*SO3-Komplex arbeiten und sehr gute Ausbeuten liefern. Für die Umsetzung von temperaturempfindlichen Substraten ist besonders die Methode mit Chloral / p-Toluolsulfon-säure geeignet. Anschließende Entschützung der Isopropylidenverbindung (±)-65 mit HCl / EtOH führte zu (±)-Wiedendiol B ((±)-2) und dem durch Gerüstumlagerung entstandenen (±)-Isowiedendiol B ((±)-66) (Abb. 5.5). Die Gesamtausbeute für (±)-Wiedendiol B ausgehend von (±)-Albicansäure ((±)-7) betrug 40 % über acht Stufen.

The present work describes the total synthesis of the sesquiterpene quinone spongiaquinone (1) and the sesquiterpene hydroquinones (±)-siphonodictyal B derivate (±)-55 and (±)-wiedendiol B ((±)-2). Spongiaquinone (1) The key intermediates for the synthesis of spongiaquinone (1) were the diastereomeric benzylic alcohols 5a,b. Starting from (-)-albicanic acid ((-)-7), (+)-albicanol ((+)-11) was synthesized by reduction of (-)-albicanic acid methyl ester ((-)-6) with DIBAL. PCC oxidation of (+)-11 gave (-)-albicanal ((-)-3), which was coupled with lithiated tetramethoxybenzene to give the diastereomeric benzylic alcohols 12a,b. Catalytic hydrogenation of 12a,b with palladium on charcoal resulted in the key intermediates 5a,b. Acetylation of 5a,b to 17a,b followed by CAN-oxidation gave the o-dimethoxybenzoquinone 19a,b. The reaction of 19a,b with p-toluene sulfonic acid yielded spongiaquinone (1). The total yield of the first total synthesis of spongiaquinone starting from (-)-albicanic acid ((-)-7) was 21 % over eight steps. The diastereomeric benzylic alcohols (±)-5a,b were transformed with trichlorotrifluoroacetone in the presence of p-toluene sulfonic acid to the 5,10-drimenylarene (±)-15, obtained by an interesting sceleton rearrangement reaction. CAN-oxidation of (±)-15 gave the o-dimethoxy-benzoquinone (±)-22. Treatment of (±)-22 with perchloric acid yielded (±)-isomamanutha-quinone ((±)-20). Siphonodictyal B derivate (±)-55 A new coupling method was used for the synthesis of the siphonodictyal B derivate (±)-55. (±)-Drimanic acid chloride ((±)-28) reacted with the appropriately protected lithiated aromatic compounds which were generated from the bromides 32, 37, 45, 56 and 13 by bromine-lithium-exchange with n-BuLi. The synthesis of the aromatic compounds could often be achieved by synthetical routes which were shorter and more effective than procedures known from literature. (±)-Drimanic acid chloride ((±)-28) was obtained in quantitative yield from (±)-drimanic acid ((±)-29) by reaction with oxalyl chloride / DMF. The ketones, generated by coupling with (±)-28, could be reduced to the corresponding diastereomeric benzylic alcohols with 100 % yield. The introduction of the formyl group in position 3' by lithiation and reaction with DMF only succeeded with styrene (±)-53, which was synthesized by reduction of ketone (±)-51 to the benzylic alcohols (±)-52a,b followed by elimination reaction (Scheme 6.4). Demethylation of the aldehyde (±)-54 with boron trichloride / tetrabutyl ammonium iodide yielded siphonodictyal B derivate (±)-55, which could not be deprotected to siphonodictyal B. (±)-Wiedendiol B ((±)-2) For the synthesis of (±)-wiedendiol B ((±)-2), the aromatic compound 57 was lithiated with t-BuLi and coupled with (±)-drimanic acid chloride ((±)-28) to give the ketone (±)-63. Reduction of the keto group with lithium triethyl boron hydride resulted in the diastereomeric benzylic alcohols (±)-64a,b. For the elimination of water from the benzylic alcohols (±)-64a,b, two new procedures were developed which work with chloral / p-toluene sulfonic acid and with pyridine*SO3-complex and produce very good yields. The procedure using chloral / p-toluene sulfonic acid is especially useful for temperature-sensitive substrates. Deprotection of the isopropylidene moiety with HCl / EtOH led to (±)-wiedendiol B ((±)-2) and (±)-isowiedendiol B ((±)-66), a byproduct produced by a carbon skeleton rearrangement (Scheme 6.5) The total yield for (±)-wiedendiol B starting from (±)-albicanic acid ((±)-7) was 40 % over eight steps.
