Abstract

Ziel dieser Arbeit war es, verschiedene, natürliche Chinone mit bereits vorhandener Antitumoraktivität zu synthetisieren und durch strukturelle Modifikationen ihre Aktivitäten oder Selektivitäten zu verbessern. Das bereits etablierte Chemotherapeutikum Doxorubicin wurde mit verschiedenen Fettsäuren und Monoterpenalkoholen verschiedener Spacerlängen via Hydrazonbindung am C-13-Kohlenstoff funktionalisiert. Zu Vergleichszwecken wurden auch Doxorubicin-Ester und ein Amid hergestellt. Es konnte gezeigt werden, dass Position C-13 und damit die Hydrazon¬bindung am besten für Funktionalisierungen geeignet ist. Konjugate mit gesättigten und ungesättigten Fettsäuren lieferten IC-50-Werte im Bereich von Doxorubicin (12 nM, 72 h, 518A2), wobei Hydrazone mit gesättigten Fettsäuren generell höhere Aktivität zeigten als welche mit ungesättigten. Als optimale Spacerlänge für Terpen-Konjugate erwies sich Dodecandisäure. Es konnte ein Menthol-Konjugat erhalten werden, welches die Aktivität von Doxorubicin mit 6 nM (IC-50, 72 h, 518A2) sogar übertraf. Das para-Benzochinon Thymochinon (TQ), welches Hauptbestandteil des Schwarzkümmels (Nigella sativa) ist, wurde an verschiedenen Positionen derivatisiert. Es zeigte sich, dass dafür der C-6-Kohlenstoff am besten geeignet ist. Es konnten mithilfe einer Radikalreaktion neben vielen Fettsäurekonjugaten auch TQ-Derivate mit end¬ständiger Carboxylgruppe unterschiedlicher Kettenlänge synthetisiert werden. Diese konnten im Nachhinein mit verschiedenen Terpenalkoholen verestert werden. Für die Fettsäurekonjugate konnte gezeigt werden, das in diesem Fall, anderes als bei Doxorubicin, mehrfach ungesättigte Fettsäuren (Linolensäure, DHA) sowie oxofunktionalisierte Fettsäuren höhere Aktivität zeigten als ihre gesättigten Vertreter. Die IC-50-Werte eines Docosahexaensäurekonjugats erreichten mit einer Konzentration von 30 nM nach 72 h sogar klinisch interessante Bereiche. Bei TQ-Terpenestern eigneten sich kurze Spacer (aus Bernsteinsäure) am besten zur Funktionalisierung. Die unterschiedlichen Terpene hatten dabei relativ wenig Einfluss auf die Aktivität. Dennoch wurde diese im Vergleich zu Thymochinon deutlich verbessert. Weiterhin konnte ausgehend von Toluolhydrochinon erfolgreich eine Synthese zur 13-C-Markierung von TQ entwickelt werden. Die Markierung wurde im Rahmen einer Grignardreaktion an einem zuvor synthetisierten Acetophenon eingeführt. Durch weitere Reaktionsschritte konnte 13-C-markiertes Thymohydrochinon erhalten werden, was mit CAN zu 13-C-TQ oxidiert wurde. Zur fluoreszenz-mikroskopischen Untersuchung von TQ und seinen Derivaten wurde dieses zusätzlich mit Biotin (für einen Streptavidin-Assay) und Alkinen („in vivo click chemistry“ mit Hilfe von Cumarinaziden) funktionalisiert. Allgemein konnte gezeigt werden, dass sich die Konjugate nach Inkubation im Zellkern befanden und dort Apoptose auslösten. Das chinoide Sesquiterpen Thespeson, welches zur Stoffklasse der Mansonone zählt, konnte erstmals erfolgreich aus einer aromatischen Komponente und einem Semiquadratsäurederivat synthetisiert werden. Die aromatische Komponente konnte dabei durch Umwandlung funktioneller Gruppen in wenigen Schritten aus 4-Methyl-2-hydroxyacetophenon hergestellt werden. Die Kontrolle über das Stereozentrum erfolgte dabei über eine wiederholte kinetisch kontrollierte enzymatische Racematspaltung mit PPL. Somit konnten beide Enantiomere mit 96 bzw. 77 % ee erhalten werden. MTT-Tests ergaben, dass natürliches (-)-Thespeson im Gegensatz zu racemischen oder (+)-Thespeson an verschiedenen Krebszellen Aktivität zeigte. Die Tatsache, dass es für gesunde Zellen vollkommen ungiftig war, macht es zu einem interessanten neuen Wirkstoffkandidaten. Auch für das Isofuranonaphthochinon 7,8-Dihydroxy-1-methylnaphtho[2,3-c]-furan-4,9-dion konnte erfolgreich eine erste Totalsynthese entwickelt werden. Zur Kontrolle der Regioselektivität wurden die beiden Edukte Veratrol und 6-Methylfuro[3,4-c]furan-1(3H)-on gewählt. Addition des Lactons an Veratrol erfolgte über ortho-Lithiierung. Oxidations- sowie Ringschluss¬reaktionen und anschließende Abspaltung der Methylgruppe führten zu dem Siderophor.

The aim of the present work was the synthesis and modification of several natural quinones already exhibiting anti-tumour activity in order to optimize their activity or selectivity. The well-known chemotherapeutic agent doxorubicin was functionalized with different fatty acids and monoterpene alcohols containing spacers of varying chain length via hydrazone bonding on the C-13 carbon. Despite of this doxorubicin-esters and a doxorubicin-amide were assembled for comparative purposes. It could be showed that the C-13 position and therefore the hydrazone bonding is optimal for functionalisation of doxorubicin. Conjugates containing saturated and unsaturated fatty acid residues exhibit IC-50-values similar to that of doxorubicin (12 nM, 72 h, 518A2). In general hydrazones with saturated fatty acids showed higher activities than those with unsaturated ones. The optimal spacer length for terpene conjugates was dodecandioic acid. One of the derivatives, the menthol conjugate, exhibited IC-50 values of 6 nM (72 h, 518A2) which even exceeded the activity of doxorubicin. The para-benzoquinone thymoquinone (TQ), which is the main constituent of black seed (Nigella sativa), was modified at different positions. It was demonstrated that the C-6 carbon is ideal for modification of TQ. By means of radical reaction many conjugates containing fatty acid residues or residues including carboxyl groups with differing spacer length could be synthesized and esterified with terpene alcohols afterwards. The fatty acid displayed, different to doxorubicin, that unsaturated (linolenic acid, DHA) or oxofunctionalisized fatty acids posses better activities than saturated ones. IC-50-values of a DHA-conjugate (30 nM, 72 h) even reached clinically interesting ranges. For thymoquinone terpene esters short spacers (three carbon atoms) seemed to be the most effective. Thereby the influence of the different terpenes on their activity was negligible. Nevertheless the efficiency in comparison to TQ could be distinctly improved. Furthermore a synthesis for 13-C labelled TQ starting from toluene hydroquinone could be developed successfully. The labelling was introduced in a beforehand synthesized acetophenone through Grignard reaction with 13-C-methyl iodide. Further functional group transformations led to thymohydroquinone which could be easily oxidised to the respective quinone using CAN. For fluorescence-microscopic analysis of TQ and its derivatives it was additionally labelled with biotin (for streptavidine assay) and acetylenes (”in vivo click chemistry“ using coumarine azides). Generally it was indicated that after incubation the TQ-derivatives were localized in the cell core and therein they induced apoptosis. The sesquiterpene quinone thespesone, belonging to the group of mansonones was synthesized successfully for the first time from two building blocks. Thereby the aromatic compound could be obtained from 4-methyl-2-hydroxy-acetophenone by functional group transformations in only a few steps. Stereo control of this system was carried out by repeated kinetic enzymatic resolution of racemic hydroxythymol using PPL. Thus both enantiomers were obtained. MTT tests showed that the natural (-)-enantiomer was far more active against various cancer cell lines including multi-resistant ones than its (+)-enantiomer and also when compared to the established antitumoral thymoquinone. The large spread between efficacy against cancer cells and tolerance by non-malignant fibroblasts makes (-)-thespesone a potential new candidate for anticancer therapy. In addition the first total synthesis for the isofuranonaphthoquinone 7,8-dihydroxy-1-methylnaphtho[2,3-c]-furan-4,9-dione could be developed successfully. In order to control regioselectivity the two building blocks veratrol and 6-methylfuro[3,4-c]furan-1(3H)-one were chosen. Addition of the lactone to veratrol was carried out by ortho-lithiiation. Oxidation, ring closure and deprotection of the catechol led to the siderophore.
