Abstract

Bet v 4 ist ein minores Allergen aus Birkenpollen (Betula verrucosa). Es gehört zur Familie der Polcalcine, kleinen Calcium bindenden Allergenen mit je 2 EF-Hand Strukturmotiven, die in einer Vielzahl höherer Pflanzen gefunden werden. Es wurden effektive Strategien zur Präparation von rekombinantem Bet v 4 entwickelt, die auch eine Markierung mit dem Isotop 15N erlaubten. Zwei hervorstechende Eigenschaft von Bet v 4 wurden bei der Reinigung ausgenutzt: Bet v 4 ist ein saures Protein, was eine Reinigung mittels Chromatofokussierung erlaubte. Aufgrund seiner Thermostabilität konnte Bet v 4 durch Kochen eines Proteinrohextraktes von hitzelabilen bakteriellen Proteinen abgetrennt werden. Somit stand genügend Bet v 4 zur Verfügung, um eine strukturelle Charakterisierung durchzuführen und letztendlich seine Struktur mittels NMR-Spektroskopie zu bestimmen. Anhand von ESI-MS, nativer PAGE und NMR-Diffusionsmessungen konnte gezeigt werden, dass Bet v 4 monomer vorliegt. Die strukturelle Charakterisierung mittels CD-Spektroskopie wies auf einen hohen Anteil alpha-helicaler Strukturelemente in der Calcium-Form hin, der anhand der chemischen Verschiebungen von alpha-Protonen, helixtypischer NOEs und vicinaler Kopplungskonstanten bestätigt wurde. Der Entzug von Calcium hat eine Verringerung des CD-Signals, jedoch keinen kompletten Verlust der Sekundärstruktur zur Folge. NMR-Spektren ließen vermuten, dass Apo-Bet v 4 keine geordnete dreidimensionale Struktur mehr besitzt. Der strukturelle Einfluss von Calcium weist mehr auf eine regulatorische Funktion von Bet v 4 als Calcium-Sensorprotein denn auf auf eine Funktion als Calcium-Speicherprotein. Durch die Auswertung von homonuklearer 2D- und heteronuklearen 3D-Spektren konnte die Struktur von Calcium gebundenem Bet v 4 gelöst werden. Die beiden EF-Hand-Motive sind durch einen flexiblen Linker miteinander verbunden und werden durch ein kurzes Faltblatt zusammengehalten. Während die beiden Calciumbindungsschleifen und die sie unmittelbar flankierenden Regionen eine starre Struktur eine aufweisen, sind die terminalen Bereiche experimentell unterbestimmt, was jedoch mit den Daten aus NMR-Relaxationsmessungen übereinstimmt, die in diesen Bereichen eine erhöhte Flexibilität nachwiesen. Der Aminoterminus ist relativ unstrukturiert. Die Kenntnis der Struktur einer Vielzahl von Allergenen wird womöglich eine Antwort auf die Frage liefern, was ein Protein zum Allergen macht.

Bet v 4 is a minor allergen from birch (Betula verrucosa) pollen. It belongs to the polcalcin family. Polcalcins are small calcium binding allergens from a wide variety of plants, which possess two calcium binding domains of EF-hand type each. In the present work effective strategies for the preparation of recombinant Bet v 4 were established, which allowed 15N-isotope labelling. Two striking properties of the birch pollen allergen were helpful for the purification of the protein: Bet v 4 is an acidic protein allowing a purification by chromatofocusing. Because of its thermostability Bet v 4 could be separated from E. coli proteins by boiling of crude bacterial lysates. A structural characterization could be carried out and finally the three-dimensional structure of Bet v 4 was determined using NMR-spectroscopy. Bet v 4 is a monomer as shown by ESI-MS, native PAGE and NMR diffusion experiments. A high content of alpha-helical structure in calcium bound Bet v 4 was detected by CD spectroscopy and was in accordance with the NMR spectroscopic data: Chemical shifts of alpha-protons, typical (i, i+3)-NOEs and vicinal coupling constants indicate Bet v 4 to predominantly possess alpha-helical secondary structure. Depletion of calcium ions leads to a decrease of the CD-signal, but not to a complete loss of secondary structure. Apo-Bet v 4 seems to lack a distinct tertiary fold as indicated by its NMR-spectra. The remarkable conformational changes upon depletion of calcium suggests Bet v 4 to be a calcium sensor protein involved in a regulatory pathway rather than a calcium buffer protein. The analysis of homonuclear two-dimensional and heteronuclear three-dimensional NMR spectra allowed the determination of the tertiary structure of calcium-bound Bet v 4. The two EF-hand motifs are connected by a flexible linker and are brought together by a short beta-sheet. The calcium binding sites and their flanking regions exhibit rigidity, while the terminal regions are underdetermined experimentally, which is consistent with NMR relaxation data showing an increased flexibility of these regions. In accordance with the relaxation data the aminoterminal amino acid residues are rather unstructured. The knowledge of the structure many of allergens might allow the answer to the question: What makes a protein an allergen?
