Abstract

Die Erdnussallergie stellt auf Grund ihrer Verbreitung und der Stärke der ausgelösten Reaktionen ein ernsthaftes Gesundheitsproblem dar. Der derzeit einzige Schutz vor den typischen Symptomen besteht in einem absoluten Verzicht auf erdnusshaltige Lebensmittel. Folglich stellt die Charakterisierung von Erdnussallergenen eine wichtige Voraussetzung für die Entwicklung von diagnostischen und therapeutischen Ansätzen dar. Als Grundlage für biochemische und biophysikalische Studien wurde eine rekombinante Expressionsstrategie zur Herstellung von authentisch gefaltetem Ara h 2 und Ara h 6 in dieser Arbeit entwickelt. Die Strategie zur effizienten Präparation großer Mengen Protein basiert auf der Anwendung spezieller E. coli Stämme mit oxidativem Cytoplasma und eignet sich zur Expression nativ gefalteter und disulfidverbrückter rekombinanter Erdnussallergene vom 2S Albumintyp und mit hoher Wahrscheinlichkeit weiterer Mitglieder dieser Proteinfamilie. Die Integrität der Sekundär- und Tertiärstruktur, der Disulfidverbrückung und der immunologischen Reaktivität der rekombinanten Proteine wurde am Beispiel von Ara h 2 mit Hilfe von natürlichem Protein aus der Erdnuss verifiziert. Die Verfügbarkeit von authentischen rekombinanten Erdnussallergenen eröffnet neue Möglichkeiten für eine verfeinerte Diagnose allergischer Erkrankungen und für die Entwicklung von spezifischen Immuntherapien. Es konnte in der vorliegenden Arbeit gezeigt werden, dass sich die Allergene Ara h 2 und Ara h 6 durch eine außerordentliche Stabilität gegenüber thermaler Denaturierung und proteolytischem Abbau auszeichnen. Durch CD spektroskopische Studien wurde eine Disulfidbrücken abhängige Stabilität der Sekundärstruktur bis 373 K nachgewiesen. Die Resistenz gegenüber Verdauungsreaktionen wurde an Hand der Enzyme Trypsin und Chymotrypsin untersucht. Der proteolytische Verdau von Ara h 2 und Ara h 6 resultierte in der Ausbildung heterodimerer immunologisch aktiver Produkte. Der proteolytische Angriff erfolgt innerhalb zweier definierter Bereiche. Die resistenten Kernbereiche der Proteine beinhalten eine Reihe von vor Proteolyse geschützten Schnittstellen. Die Faltungen der unverdauten Proteine und der proteaseresistenten Kernbereiche sind nahezu identisch. Die proteaselabilen Bereiche weisen keine reguläre Sekundärstruktur auf und sind von einer signifikanten Flexibilität geprägt. Die für die Erdnussallergene Ara h 2 und Ara h 6 nachgewiesene Stabilität stellt ein wichtiges Merkmal der Lebensmittelallergene dar. Je länger ein möglichst großer Proteinabschnitt intakt bleibt, umso höher ist die Wahrscheinlichkeit, dass eine Immunantwort ausgelöst werden kann und umso stärker wird diese ausfallen. In der vorliegenden Arbeit konnte die Lösungsstruktur des proteaseresistenten Ara h 6-Kerns bestimmt und charakterisiert werden. Die resultierende Struktur besteht aus fünf alpha-Helices, die einen vor enzymatischer Spaltung geschützten kompakten Kernbereich ausbilden. Die ermittelte Struktur stellt die erste experimentell bestimmte dreidimensionale Struktur eines Erdnussallergens und die erste hochaufgelöste Struktur eines 2S Albumins dar. Die globale Faltung von Ara h 6 aus der Erdnuss ähnelt weiteren Pflanzenproteinen: den nichtspezifischen Lipidtransferproteinen, den Amylase/Trypsin-Inhibitoren und dem hydrophoben Protein aus Sojabohne. Eine wichtige Frage betrifft die Lokalisation der IgE bindenden Epitope. In früheren Studien konnten in Ara h 6 keine linearen IgE reaktiven Epitope identifiziert werden, für Ara h 2 wurden hingegen drei immunodominante Regionen beschrieben. Um die Frage nach der Lokalisation dieser Epitope innerhalb der dreidimensionalen Struktur von Ara h 2 zu klären, wurde ein molekulares Modell des Proteins auf der Basis der experimentell gelösten Ara h 6 Struktur erstellt. Sämtliche Epitope befinden sich in einem ausgedehnten Schleifenbereich und am aminoterminalen Ende des Proteins. Dieser Befund weist in Kombination mit der Kenntnis, dass die Multivalenz eines Allergens eine Voraussetzung für das Auslösen einer allergischen Antwort darstellt und mit ihrer Intensität korreliert, stark auf die Existenz von konformationellen Epitopen in Ara h 6 und Ara h 2 hin. Die Präsentation von stabilen konformationellen Epitopen auf Mastzellen führt im Allgemeinen zu einer effizienteren und stärkeren Mediatorausschüttung. Dies ist mit den beobachteten starken klinischen Symptomen bei Patienten mit Erdnussallergie konsistent. Zusammenfassend kann festgestellt werden, dass die Stabilität und Kompaktheit der Struktur sowie das potentielle Vorhandensein von konformationellen Epitopen eine Erklärung für die extreme Allergenizität der Erdnussallergene liefern kann. Die Identifizierung von IgE-bindenden Epitopen auf der Basis der dreidimensionalen Struktur der Erdnussallergene liefert eine Grundlage für die Entwicklung von veränderten Proteinen mit reduzierter Allergenizität, dem Verständnis von Kreuzallergien und der Entwicklung neuer Therapieformen.

Peanut allergy is a significant health problem because of the prevalence and severity of the reaction. The only therapeutic option currently available for the prevention of a peanut hypersensitivity reaction is food avoidance. Therefore, the characterization of peanut allergens is important for developing diagnostic and therapeutic capabilities. As a basis for the biochemical and biophysical studies of the two allergenic proteins, a recombinant expression strategy of authentically folded Ara h 2 and Ara h 6 was established in this work. This efficient large-scale method exploiting on special E. coli strains with an oxidizing cytoplasm is clearly suited for recombinant expression of properly folded and disulfide bridged peanut 2S albumins and highly likely other members of this protein family. The intact secondary and tertiary structure, correct disulfide pattern and immunological reactivity of the recombinant proteins was confirmed using the native Ara h 2 protein from peanut as a reference. The availability of authentic recombinant peanut allergens presented in this work may open new opportunities for refined allergic diagnosis and the development of specific immunotherapies. In the present work first biophysical studies focused on the thermal and proteolytic stability of Ara h 6 and Ara h 2. It could be shown that Ara h 2 and Ara h 6 are extremely stable proteins resistant to thermal denaturation and to proteolytic digestion. CD spectroscopic studies showed clearly that the secondary structure of Ara h 2 and Ara h 6 is stable until 373 K. This thermal stability is lost in the presence of a reduction agent which indicates that it is mediated by the disulfide bridge connections. Resistance to digestion was demonstrated using trypsin and chymotrypsin. The protease-resistant cores of Ara h 2 and Ara h 6 were characterized. The digestion results in the creation of heterodimeric proteins that could be shown as immunological active. Aminoterminal sequencing of the subunits revealed that digestion occurs at two distinct regions in the proteins. The resistant protein cores containing several potential enzyme cleavage sites are protected from digestion. The folding of the protease-resistant cores of Ara h 2 and Ara h 6 is nearly identical to the undigested protein. The protease-labile parts are regions without regular secondary structure and with a significant flexibility. The stable nature of proteins is thought to be an important characteristic of food allergens because the longer significant portions of the protein remain intact the more likely it is to trigger an immune response. Furthermore, in the present work the solution structure of the protease-resistant Ara h 6 core has been determined by NMR spectroscopy. The resulting compact structure consists of a right handed superhelix formed by five alpha-helices. This structure represents the first experimentally solved threedimensional structure of a peanut allergen and the first high resolution structure of a 2S albumin. The global fold of Ara h 6 from peanut is similar to that of other classes of plant proteins: nonspecific lipid transfer proteins, amylase/trypsin-inhibitors and the hydrophobic protein from soybean. One major question concerns to the localization of epitopes. Whereas no linear IgE reactive epitopes were identified in Ara h 6, three immunodominant regions were defined in Ara h 2 in earlier studies. The question, where these linear epitopes are located in the three-dimensional structure was addressed by building a molecular model of Ara h 2 on the basis of the experimentally solved structure of Ara h 6. All epitopes are clustered in an extended loop and in the aminoterminal area of the protein. As it could be shown in this work both regions are highly sensitive to proteolytic digestion. These findings, in combination with the knowledge that allergens must be multivalent to elicit an allergic response, point strongly toward the existence of conformational epitopes in Ara h 6 and Ara h 2 as well. The presentation of stable, conformational epitopes to mast cells generally results in a more efficient and dramatic release of mediators, consistent with severe clinical symptoms observed in patients with peanut hypersensitivity. Taken in total, the very stable nature of the compact structure and the likely existence of conformational epitopes offers an explanation for the extreme allergenicity of these peanut allergens. Identification of IgE-binding epitopes on the basis of the three-dimensional structures of these peanut allergens provides the basis for the development of altered proteins with reduced allergenicity, the explanation of cross reactiveness and for novel designs of allergy treatment.
