Abstract

Viele Proteine in der Zellmembran oder im Blutserum sind in eukaryotischen Organismen glycosyliert. Da die Kohlenhydratstrukturen verschiedene Aufgaben erfüllen, ist die Zugänglichkeit uniformer Glycoproteine für die Untersuchung von Struktur-Wirkungsbeziehungen nötig. Aufgrund der Mikroheterogenität ist die Isolierung von N-Glycanen aus Glycoproteinen erschwert. Daher stellt die chemische Synthese eine gute Alternative dar. Relativ selten in der Natur kommen N-Glycane des bisecting-Typs vor, deren Synthese in der vorliegenden Arbeit beschrieben wird. Diese Verbindungen sollen für die Untersuchung von Wechselwirkungen mit Lectinen, Seren oder Antikörpern auf Glycochips bereitgestellt werden. Substituierte N-Glycane können aus modularen Bausteinen hergestellt werden, aus denen auch ein pentaantennäres N-Glycan mit bisecting-GlcNAc und Core-Fucose synthetisiert werden sollte. Hierbei zeigte sich, dass die Ausbeute der sterisch anspruchsvollen Glycosylierung des hoch verzweigten alpha-1,6-Arms an den Heptasaccharidakzeptor von zwei Schlüsselschutzgruppen abhängt. Bei der Verwendung von zwei verschiedenen Donorbausteinen, bei denen die peripheren Aminogruppen durch Phthalimidoschutzgruppen geschützt wurden, wirkte sich das Fehlen der 3-Acetatschutzgruppe an der Mannose positiv auf die Ausbeuten aus. Einen ähnlichen Effekt hatte der Austausch der sperrigen Phthalimidoschutzgruppen gegen kleinere Trifluoracetamide in den Donoren. Mit diesen Bausteinen konnte die Ausbeute weiter verbessert werden, was auf elektronischen und sterischen Effekten beruhen sollte. Nach der Optimierung der Glycosylierung zu pentaantenären Strukturen wurde eine Core-Fucose eingeführt und die basenlabilen Schutzgruppen wurden entfernt. Damit konnte eine vielseitige Einsetzbarkeit der modularen Bausteine mit der Trifluoracetamidschutzgruppe gezeigt werden. Aufgrund dieser Ergebnisse wurden neue modulare Bausteine entwickelt, um die Synthese der verschieden verzweigten N-Glycane und die nachträgliche Einführung eines bisecting-GlcNAc-Rests zu untersuchen. Durch die unterschiedliche Kombination der neuen Antennendonoren mit dem Core-Trisaccharid konnten die komplexen N-Glycane mit zwei bis vier Antennen hergestellt werden. Für die Glycosylierungsreaktionen mussten aufgrund der erhöhten Reaktivität der trifluoracetamidgeschützten Donorbausteine geeignete Reaktionsbedingungen ermittelt werden. Die Regio- und Stereoselektivität in diesen Reaktionen wurde über die Konzentration und Temperatur der Reaktionsansätze gesteuert und ungewollte Überreaktion an benachbarten Hydroxylgruppen konnte damit unterdrückt werden. Mit Hilfe des GlcNAc-Donors konnte der bisecting-Rest nachträglich in die N-Glycane eingeführt werden. Durch die modularen trifluoracetamidgeschützten Bausteine wurden N-Glycane mit und ohne bisecting-GlcNAc in einer Syntheseroute erhältlich und die bisherige Verwendung einer temporären Chloracetatschutzgruppe unnötig. Es wurde zusätzlich die sequentielle Einführung des alpha-1,6-Arms und des bisecting-GlcNAc-Rests in regio- und stereoselektiven Eintopfreaktionen gezeigt. Auf diese Weise konnten auch das komplexe pentaantennäre N-Glycan und ein N-Glycan des Hybridtyps mit bisecting-Substitution hergestellt werden. Die vier chemisch synthetisierten N-Glycane des bisecting-Typs wurden in sechs Reaktionsschritten entschützt, an den bifunktionalen Spacer für die Immobilisierung auf Glycochips geknüpft und an den terminalen N-Acetylglucosaminyleinheiten mit Glycosyltransferasen enzymatisch verlängert. Hierbei wurden zuerst Galactosen beta-1,4-glycosidisch angeknüpft, an welche Sialinsäuren alpha-2,3- oder alpha-2,6-glycosidisch gebunden werden konnten. Somit wurden für die Immobilisierung auf den N-Glycanchips erstmals 17 komplexe N-Glycane mit bisecting-GlcNAc zur Verfügung gestellt. Außerdem wurde in weiteren Versuchen die Kopplung von N-Glycanen an die Seitenkette des Asparagins untersucht. Hierzu wurde das Core-Trisaccharid nach Reduktion des Azids an ein Asparaginsäureamid geknüpft und anschließend entschützt. Außerdem konnte das Glycosylhydroxylamin selektiv über eine decarboxylative Kondensation an ein Aminosäurederivat gebunden werden.

Many proteins of the cell membrane or serum are glycosylated in eukaryotic organisms. Since the carbohydrate structures fulfill different functions, the availability of uniform glycoproteins for structure-activity relationships is necessary. The microheterogeneity complicates the isolation of N-glycans from glycoproteins. Thus chemical synthesis is a good alternative. Relatively rare in nature are N-glycans of the bisecting type, whose synthesis is described in the present thesis. These compounds should be provided for the analysis of interactions with lectins, sera or antibodies on glycoarrays. Branched N-glycans can be synthesized with modular building blocks, from which a pentaantennary N-glycan with bisecting GlcNAc and core fucose should be synthesized. It was shown that the yield in the sterically demanding glycosylation of the highly branched alpha-1,6-arm and the heptasaccharide acceptor depends on two key protection groups. When using two different donor building blocks with phthalimido protecting groups for the peripheral amino functions the lack of an acetate at position 3 of the mannose influenced the yield positively. The exchange of the bulky phthalimido groups for the smaller trifluoroacetamides in the donors showed a similar effect. With these building blocks the yield could be further improved, which should be related to electronic and steric effects. After optimization of the glycosylation reaction leading to pentaantennary structures, a core fucose was added and the base labile protecting groups were removed. These results show a several applicability of the modular building blocks with trifluoroacetamide protecting groups. Because of these results new modular building blocks were developed in order to test the synthesis of differently substituted N-glycans and the subsequent introduction of a bisecting GlcNAc residue. By a combination of the new antennae donors with the core trisaccharide the complex N-glycans with two to four antennae were synthesized. Because of the increased reactivity of the trifluoroacetamide protected donors, adequate conditions had to be found for the glycosylation reactions. Regio- and stereoselectivity of these reactions was affected by the concentration and temperature of the reactions and unintentional overreaction at neighboring hydroxyl groups could be reduced. The bisecting residue could be introduced subsequently into the N-glycans via a GlcNAc-donor. N-glycans with and without bisecting GlcNAc could be obtained by the modular trifluoroacetamide protected building blocks following a single route and the previous use of a temporary protecting group became unnecessary. The sequential introduction of alpha-1,6-arm and bisecting GlcNAc were additionally shown in a region- and stereoselective one pot glycosylation. In this way the complex pentaantennary N-glycan and a hybrid type N-glycan with bisecting substitution could be obtained. The four chemically synthesized N-glycans were deprotected in six reactions, coupled to a bifunctional linker for the immobilization on glycochips and enzymatically elongated at the terminal N-acetylglucosamine residues by glycosyltransferases. The first elongation attached beta-1,4-glycosidically linked galactose, followed by an alpha-2,3- or alpha-2,6-sialylation. For the first time 17 complex N-glycans with a bisecting GlcNAc are provided for the immobilization on N-glycan arrays. Furthermore the coupling of N-glycans to the side chain of asparagines was investigated. Therefore the core trisaccharide was attached to an aspartic amide after reduction of the azide followed by deprotection. Additionally the glycosylhydroxylamine was converted to a glycosylaminoacid by decarboxylative condensation with an alpha ketoacid.
