Abstract

Mit Pt(II)-Komplexen der 6’-Aminomethylnicotinsäure konnte ein neues Metallkomplexfragment entwickelt werden, über das potentielle Wirkstoffe über wenige Syntheseschritte verbunden werden konnten. Eine erste erfolgreiche Entwicklung war ein (-)-Menthylester-Konjugat mit Potential zur Überwindung der problematischen Cisplatin-Resistenz bei Hodenkrebszellen. Anschließende Modifikationen der Terpen- und der Ligand-Komponente führten zu einem (+)-Cedrenyl-Derivat mit verbesserter Wirkung bei Cisplatin-resistenten Melanom- und Hodenkrebszellen, zu Menthol-Konjugaten mit Diaminopropionat- und -butyrat-Liganden und höherer Aktivität bei Darmkrebszellen, sowie zu Propyl-Spacer-verknüpften Menthol- und Neomenthol-Konjugaten mit ebenfalls verbesserter Wirkung bei diversen resistenten Krebsformen. Zur Erhöhung der Selektivität von Pt(II)-Verbindungen auf hormonabhängige Krebsarten wie Brustkrebs wurde in dieser Arbeit ihre Bindung an das Serumprotein SHBG optimiert, welches Estrogene und Androgene im Blut transportiert und im Zielgewebe akkumuliert. Eine erste Serie von Steroid-Konjugaten mit Pt(II)-Komplexen wies deutliche Struktur-Aktivitäts-Beziehungen hinsichtlich der Affinität zu SHBG auf. Vor allem über die 3-Hydroxy-Gruppe veresterte Estradiol- und Estron-Konjugate sowie ein über den D-Ring verknüpftes Testosteron-Konjugat zeigten eine starke Bindung an SHBG. Ru(Aren)-Komplexe besitzen in vielerlei Hinsicht Vorteile gegenüber Platinverbindungen. So führte die einfache Synthese von entsprechenden ternären Konjugat-Komplexen unter milden Bedingungen ausgehend von einem Ru(Aren)-Dimer zu Konjugaten mit dem Phase-III-Wirkstoff Abirateron, dem stark auf Nierenkrebs wirkenden Digitoxigenin, oder zwei Rezeptortyrosinkinase-inhibierenden Tyrphostin-Derivaten. Ein weiteres Projekt war die Synthese von Metallkomplexen mit Melophlinen, einer Familie mariner N-Methyl-3-acyltetramsäuren. Melophlin C wurde mit geeigneten Metall-Fragmenten zu den entsprechenden Metallkomplexen umgesetzt, wobei La(III)- und Ru(II)-Komplexe eine starke Wirkung auf Nierenkrebs- (A-498) und Leukämiezellen (U-937) im niedrigen mikro- bis sub-mikromolaren Bereich zeigten. Der giftige Pilzinhaltsstoff Illudin-M wurde aus Flüssigkulturen von Omphalotus olearius isoliert und derivatisiert. Dimer-Verbindungen, in welchen Illudin M mit Bipyridyldicarbonsäure oder mit Endothall über die Yamaguchi-Methode verestert wurde, zeigten eine erhöhte Wirkung auf Bauchspeicheldrüsenkrebs und eine verringerte Toxizität bei nicht-malignen Hautfibroblastenzellen. Nachfolgende Arbeiten führten zur einem Phthalester, der die oben genannten Diester in der Wirkung übertraf. Der dritte Themenkomplex dieser Arbeit befasste sich mit Derivaten des Phase-III-Wirkstoffs Combretastatin A-4 und dem Ziel, die Effizienz sowie die pharmakologischen Eigenschaften zu verbessern. Zur Konjugatsynthese eigneten sich in erster Linie bekannte Chalkon-Analoga. Bemerkenswert ist vor allem ein neues Pt-Konjugat, welches bei resistenten Tumorarten die Wirkung des freien Chalkons übertraf. Eine wertvolle Alternative zu den Chalkonen stellte ein hochwirksames Oxazol-verbrücktes Analogon dar, welches über die van-Leusen-Reaktion erhalten werden konnte. Ein Ru(II)-Konjugat, welches aus dem Isonicotinsäureester des Oxazols und einem Ru-Dimer hergestellt wurde, erzielte bei einem Darmkrebs mit WT-p53 eine verbesserte Wirkung und scheint durch die Ru(II)-Komponente einen zusätzlichen Wirkmechanismus zu besitzen, der möglicherweise über eine Schädigung der DNA verläuft. Außerdem wurden weitere Oxazol-Derivate synthetisiert. Ein Methylsulfanyl-Derivat konnte die Combretastatin-Resistenz des HT-29-Darmkrebses überwinden. Ein Oxazol-verbrücktes Analogon von Combretastatin A-1, welches eine p53-abhängige Wirkung zu besitzen scheint, konnte zu einem p-Chinon oxidiert werden. Abschließend wurde eine der meta-Methoxygruppen des 3,4,5-Trimethoxyphenyl-Rings gegen Halogene (Cl, Br) oder eine Amino-Gruppe ersetzt, und unterschiedlichste Oxazol- und N-Methylimidazol-verbrückte Derivate mit dieser Modifizierung hergestellt. Durch Überführung in HCl-Salze konnten wasserlösliche Vertreter mit ansprechender Antitumor-Aktivität erhalten werden, die für eine in-vivo-Anwendung in Testmäusen besonders geeignet sind.

A new Pt(II) complex fragment based on 6-aminomethylnicotinic acid was developed, and potential co-drugs could be easily connected to this moiety via few synthetic steps. A first promising derivative was a novel (-)-menthyl ester conjugate with potential to overcome cisplatin resistance in testicular cancer cells. Subsequent modifications of the terpene and the N,N-ligand moieties led to the (+)-cedrenyl conjugate with better impact on cisplatin resistant melanoma and testicular cancers, as well as to menthol conjugates featuring diaminopropionate and –butyrate ligand systems exhibiting enhanced growth inhibitory effects on colon cancer cells, and to propyl-spacered menthol and neomenthol conjugates with higher activity in various resistent tumour cell lines. In order to enhance the selectivity of Pt(II) based drugs for hormone dependent cancer types such as breast cancer their binding affinity to the serum protein SHBG was optimised. This protein regulates the blood transport of steroidal estrogens and androgens, and accumulates in the target organs. A first series of steroid conjugates with dichloridoplatinum(II), prepared analogously to the synthesis of the terpene conjugates, revealed marked structure SHBG affinity relationships. In particular, the estradiol and estrone conjugates linked via the 3-hydroxy group as well as a testosterone conjugate connected via the D-ring to the Pt(II) moiety showed excellent binding to SHBG. Ru(II) complexes offer some advantages over Pt(II)-based drugs. The synthesis of ternary Ru(II) complexes is straightforward and proceeds under mild conditions starting from a Ru(arene) dimer. In this way, conjugates with the phase-III-drug abiraterone, with highly active digitoxigenin and with RTK inhibiting tyrphostine derivatives were obtained. Another project dealt with the synthesis of metal complexes of melophlins, a family of marine N-methyl-3-acyltetramic acids. The cytotoxic derivative melophlin C was treated with suitable metal sources to give the corresponding metal complexes. Here, La(III) and Ru(II) complexes exhibited distinct activities on renal cancer (A-498) and leukemia cells (U-937) in the low micro and sub-micro molar range. The natural spirocyclopropane illudin M, a highly cytotoxic sesquiterpene produced by Omphalotus olearius, was isolated from culture liquids and was used as a starting material for the synthesis of less toxic derivatives. Dimeric compounds featuring bipyridyldicarboxylic acid or endothall esterified to illudin M showed an enhanced impact on pancreatic cancer cells when compared to free illudin M while being less toxic in non-malignant skin fibroblast cells. Subsequent work led to the discovery of a phthalic ester, reaching or even exceeding the anticancer activity of the previous diesters. The third part of this thesis describes various derivatives of the cytotoxic and vascular-disrupting agent combretastatin A-4. They are aimed at an improvement in efficiency and in the pharmacological properties compared to the lead compound. Some already known chalcone analogues turned out to be very suitable as part of active conjugates. A Pt(II) conjugate, prepared analogously to the synthesis of the terpene-Pt-conjugates, could surpass the growth inhibiting effect of the parent chalcone on various resistant tumours. A known oxazole-bridged combretastatin analogue available via the van-Leusen reaction was found to be a valuable alternative to the chalcones mentioned above. A conjugate of this 4,5-diaryl oxazole with a dichlorido(cymene)Ru moiety exhibited a higher activity than the parent oxazole in colon cancer cells with intact p53 protein and seemed to have an additional cytotoxic mode of action possibly working via ruthenation of DNA or proteins. In addition, the parent oxazole itself was further modified in order to obtain more active and better water soluble drug candidates. For instance, a methylsulfanyl derivative broke the combretastatin A-4 resistance of the colon cancer HT-29. An oxazole-bridged analogue of the catechole derivative combretastatin A-1 was likewise synthesized. It revealed a p53 dependent activity and underwent oxidation to a p-quinone in aqueous medium containing Fe(III). Finally, one of the meta-methoxy groups of the combretastatin 3,4,5-trimethoxyphenyl ring was replaced by halogens (Cl, Br) or an amino group, and various oxazole- and N-methylimidazole-bridged derivatives were prepared having this modification. By conversion into the corresponding hydrochloride salts water soluble agents were available which are appropriate for an in vivo application in laboratory animals.
