Abstract

Die Arbeit beschäftigt sich mit der stereoselektiven Totalsynthese von biologisch aktiven Alkaloiden und natürlich vorkommenden Sesquiterpenchinonen. Dabei wurden bewusst Beispiele ausgewählt, die aufgrund ihrer biologischen Funktion, pharmakologischen Wirkung oder unklarer Stereochemie besonders interessant sind. Im ersten Teilprojekt sollten gezielt Isomere von 5-(2-Methylbutyliden)-2,3,4,5-tetrahydropyridin, dem gemeinsamen biosynthetischen Vorläufer der antibakteriell und gegenüber Prädatoren abschreckend wirkenden Stenus-Alkaloide Stenusin, Cicindeloin und 3-(2-Methyl-1-butenyl)-pyridin dargestellt werden. Nach Synthese von drei der vier möglichen Isomere konnte durch GC/MS-Vergleich von synthetisierten und natürlichen Alkaloiden gezeigt werden, dass das (S,E)-Isomer in Stenus similis und Stenus cicindeloides und das (R,E)-Isomer in Stenus tarsalis vorkommt. Dadurch konnte die postulierte Struktur der biosynthetischen Zwischenstufe der Stenus-Alkaloide eindeutig bestätigt werden. Ein weiteres Teilprojekt bestand in der stereoselektiven Synthese zweier im Arbeitskreis Seifert aus der Glasperlenanemone Heteractis aurora isolierten Aplysinopsin-Alkaloide, um deren Absolutkonfiguration zu bestimmen. Diese Klasse der Indolalkaloide besitzt pharmakologische Bedeutung als potentielle neue MAO-Hemmer und 5-HT2-Agonisten. Dazu wurde zur Darstellung der einfacheren Verbindung von natürlichem L-Tryptophan ausgegangen. Für den bromierten Naturstoff wurde racemisches N-Acetyl-6-brom-DL-tryptophan über eine Leimgruber-Batcho-Indolsynthese und anschließende Kondensation mit L-Serin dargestellt und dieses durch enzymatische Racematspaltung in 6-Brom-L-tryptophan überführt. Aus beiden Aminosäuren wurden in drei bzw. vier Stufen die gewünschten Naturstoffe dargestellt. Dabei trat bei beiden Verbindungen in der letzten Stufe teilweise Racemisierung auf. Die negativen optischen Restrotationen deuten aber darauf hin, dass beide rechtsdrehenden Naturstoffe (R)-Konfiguration besitzen sollten. Im Hauptteil der Arbeit beschäftigte ich mich mit der erstmaligen asymmetrischen Synthese der natürlich vorkommenden Sesquiterpenhydrochinone bzw. -chinone Deoxyspongiachinol, Deoxspongiachinon, 3-Oxo- und 3β-Hydroxytauranin. Derartige Verbindungen besitzen meist vielfältige biologische Wirkungen wie z.B. antibakterielle, antivirale, cytotoxische oder antileukämische Aktivität. Die Synthese der antibakteriell wirkenden Verbindungen Deoxyspongiachinol und –chinon wurde gezielt so geplant, dass dabei die interessanten Naturstoffe und wertvollen Synthesebausteine (+)-Albicanol, (+)-Drimanol und (+)-Drimansäure durchlaufen werden. Dazu wurde aus Geraniol hergestelltes all-trans-Farnesylacetat unter Verwendung des Noe-Lin-Liganden, dessen Herstellung im Rahmen dieser Arbeit verbessert wurde, asymmetrisch dihydroxyliert. Das erhaltene Glykol wurde zum Epoxid umgesetzt und mittels einer Ti(III)-katalysierten, bioinspirierten Cyclisierungskaskade in (+)-3β-Hydroxyalbicanylacetat überführt. Nach optimierter Barton-McCombie-Desoxygenierung und Deacetylierung zu (+)-Albicanol wurde ein neues, (PPh3)3RuCl2 katalysiertes Verfahren zur hoch diastereoselektiven Hydrierung (de =98%) zu (+)-Drimanol entwickelt. Nach direkter PCC katalysierter Oxidation mit H5IO6 zur (+)-Drimansäure konnte diese in insgesamt neun Stufen in einer Gesamtausbeute von 13,3% aus all-trans-Farnesol hergestellt werden. Das benötigte Arylbromid wurde aus Vanillin durch Bromierung, Dakin-Oxidation und anschließender doppelter MEM-Schützung über drei Stufen in 64,4% Ausbeute erhalten. Dieses wurde nach Lithiierung unter optimierten Bedingungen mit dem aus (+)-Drimansäure hergestellten Drimanoylchlorid zum (–)-Drimanophenon gekuppelt. Reduktion, Eliminierung und Entschützung lieferte Deoxyspongiachinol über zwei Stufen in sehr guter Ausbeute (78,4% bezogen auf das Drimanophenon). Im abschließenden Schritt konnte dieses mit Ammoniumcer(IV)-nitrat annähernd quantitativ zu Deoxyspongiachinon oxidiert werden. Somit konnten beide natürlichen Meroterpene in sehr guter Ausbeute über einen bioinspirierten Zugang gewonnen werden und durch Vergleich der Drehwerte deren Absolutkonfiguration zu (5S,8S,10S) festgelegt werden. Bei der Darstellung von 3-Oxo- und 3β-Hydroxytauranin wurde eine Strategie entwickelt, die die selektive Oxidation eines 3β-Hydroxytauranin-Derivats zu einem 3-Oxotauranin-Derivat erlaubt und somit durch die späte Verzweigung des Syntheseweges viele Stufen im Vergleich zu einer separaten Darstellung beider Verbindungen einspart. Einige Schutzgruppenkonfigurationen am Arylbaustein führten zwar ins Leere, allerdings wurde dabei eine bis dato unbekannte Möglichkeit gefunden, um TIPS-MEM geschützte Verbindungen durch die Wahl der Natur des nukleophilen Lösungsmittels selektiv zu entschützen. Für die letztendlich zielführende Strategie wurde (E)-6,10-Dimethylundeca-5,9-dien-1-in benötigt. Alle bis dato literaturbekannten Routen benötigen dafür entweder gasförmiges Allen oder Propin oder bedienten sich einer reduktiven Entschweflung von (E)-(6,10-Dimethylundeca-5,9-dien-1-in-3-yl)(phenyl)sulfan unter streng wasserfreien Birch-Bedingungen. Im Rahmen dieser Arbeit wurde für letztere Reaktion eine gut handhabbare und leicht skalierbare (> 100 mmol) Alternative entwickelt, die katalytische Mengen an Naphthalin und einen Überschuss an Lithiumstaub verwendet. Das zur geplanten Negishi-Kupplung benötigte Benzylchlorid wurde dabei über sieben Stufen aus 3,5-Dihydroxybenzoesäure in einer exzellenten Gesamtausbeute von 56,8% dargestellt. Dabei wurde ein Verfahren entwickelt, das praktisch beliebig skalierbar ist (hier: > 40 g Benzylchlorid in einem Durchgang dargestellt), da es nur preiswerte Reagenzien verwendet und auf Umkristallisation zur Aufreinigung beruht. Nach Carboaluminierung des Alkins und darauf folgender Negishi-Kupplung mit dem Benzylchlorid konnte nach zahlreichen Optimierungsversuchen das gewünschte Farnesylaren in guter Ausbeute (83%) erhalten werden. Diese Verbindung wurde nach vielen Optimierungen der Reaktionsbedingungen mit dem Noe-Lin-Liganden selektiv dihydroxyliert. Nach Umsetzung zum Epoxid, bioinspirierter Domino-Cylisierung und Debenzylierung stand das entsprechende Resorcinderivat zur Verfügung. Nach Oxidation zum 2-Hydroxychinon, für die nach umfangreichen Studien Phenyliod(III)bis(trifluoracetat) als optimales Reagenz (91% Ausbeute) ermittelt wurde, und Desilylierung wurde der gewünschte Naturstoff 3β-Hydroxytauranin erhalten. Selektive Oxidation der sekundären OH-Gruppe mit Dess-Martin-Periodinan unter streng kontrollierten Bedingungen neben der sehr oxidationsempfindlichen 2-Hydroxychinon-Einheit an der noch silylgeschützen Verbindung liefert nach Desilylierung 3-Oxotauranin. Alle spektroskopischen Daten stimmen sehr gut mit denen für die beiden Naturstoffe publizierten Werten überein. Somit konnten beide Verbindungen erstmalig totalsynthetisch dargestellt und zudem gezeigt werden, dass es sich beim Naturstoff um 3β-Hydroxytauranin und nicht wie postuliert um 3α-Hydroxytauranin handelt.

This thesis deals with the stereoselective total synthesis of biological active alkaloids and natural occurring sesquiterpene quinones. The consciously chosen examples are very interesting because of their biological function, pharmacological activity or uncertain stereochemistry. In the first subproject isomers of 5-(2-methylbutylidene)-2,3,4,5-tetrahydropyridine should be synthesized in a stereospecific manner. This compound is the common biosynthetic precursor of the Stenus alkaloids stenusine, cicindeloine and 3-(2-methyl-1-butenyl)-pyridine, which possess antibacterial properties and deterrent activities against predators. By synthesis of three of the four possible isomers it was shown that Stenus similis und Stenus cicindeloides contain the (S,E)-isomer and the (R,E)-Isomer occurs in Stenus tarsalis. This was proved by comparison of GC/MS data of the prepared compounds with samples from the corresponding natural sources. As a result the postulated structure of the common biosynthetic precursor of the Stenus alkaloids could be validated. Another subproject was the stereoselective synthesis of two indole alkaloids to determine their absolute configurations. Both compounds have been isolated in the Seifert group from the sea anemone Heteractis aurora and belong to the group of aplysinopsines showing interesting pharmaceutical activities like MAO-inhibition or 5-HT2-agonism. The preparation of the simpler compound started with natural L-tryptophan. For the preparation of the brominated alkaloid racemic N-acetyl-6-bromo-DL-tryptophan was obtained by a Leimgruber-Batcho indole synthesis and subsequent condensation with L-serine. Aminoacylase mediated enzymatic resolution of the so obtained racemic mixture resulted in 6-bromo-L-tryptophan. Both amino acids were converted in three respectively four steps to the desired alkaloids. Although racemization was a serious issue during the last step in the synthesis of both alkaloids, residual negative optical rotations could be measured for both synthetic compounds in contrast to the positive rotations of the natural products. So both natural products should possess (R)-configuration. The main focus of this thesis is the enantio- and diastereoselective synthesis of the sesqui¬terpene quinones and –hydroquinones deoxyspongiaquinol, deoxspongiaquinone, 3-oxo- und 3β-hydroxytauranin. Such compounds often have diverse biological effects e.g. antibacterial, antiviral, cytotoxic or antileukemic activity. The synthesis of deoxyspongiaquinol and deoxyspongiaquinone was intentionally designed to entrain the interesting natural occurring sesquiterpenes (+)-albicanol, (+)-drimanol, and (+)-drimanic acid, which are precious building blocks for the synthesis of other interesting meroterpenes. All-trans-farnesyl acetate derived from geraniol was dihydroxylated using the Noe-Lin ligand, which preparation was improved in course of this work. The obtained glycol was converted to the corresponding oxirane and then subjected to a Ti(III)-promoted, bioinspired cyclization cascade to furnish 3β-hydroxyalbicanyl acetate. An optimized Barton-McCombie deoxygenation followed by methanolic cleavage of the acetate moiety led to (+)-albicanol in excellent yield. Subsequently, a highly diastereoselective (de = 98%) hydrogenation protocol utilizing (PPh3)3RuCl2 to yield (+)-drimanol was developed. This primary alcohol was directly oxidized to (+)-drimanic acid in almost quantitative yield using catalytic amounts of PCC and H5IO6 as terminal oxidant. So (+)-drimanic acid was synthesized from all-trans-farnesol in nine steps with an overall yield of 13.3%. The required aryl bromide was prepared from vanillin via regioselective bromination, Dakin oxidation and MEM protection over three steps in 64.4% yield. This was lithiated and coupled under optimized conditions with drimanoyl chloride derived from (+)-drimanic acid to furnish the corresponding (–)-drimanophenone. Reduction, elimination and deprotection provided (+)-deoxyspongiaquinol over two steps in very good yield (74.8% based on the drimanophenone). A final oxidation with ceric ammonium nitrate provided (+)-deoxyspongiaquinone as a further natural product in almost quantitative yield. Consequently, both natural meroterpenes could be prepared in very good yield via a bioinspired approach and the absolute configuration of both natural products could be established as 5S,8S,10S. For the synthesis of the two other meroterpenes a straightforward stereoselective approach to 3β-hydroxytauranin, allowing a late stage oxidation to 3-oxotauranin, should be developed. Also some of the tested protecting group strategies weren´t successful, it should be noted, that a novel protocol for the highly selective deprotection of TIPS-MEM protected compounds was found. The protecting group to be removed can be chosen by the nature of the nucleophilic solvent. For the finally successful strategy in the synthesis of the tauranins (E)-6,10-dimethylundeca-5,9-dien-1-yne was required. All known approaches use either gaseous allene or propyne or utilize a reductive desulfurization of (E)-(6,10-dimethylundeca-5,9-dien-1-yn-3-yl)(phenyl)sulfane under strictly anhydrous Birch conditions. In course of this work a novel, scalable (> 100 mmol) and more convenient alternative for the latter reaction using catalytic amounts of naphthalene and an excess of lithium dust was developed. The benzyl chloride required for the pending Negishi coupling was prepared from 3,5-dihydroxybenzoic acid over seven steps in an excellent overall yield of 56.8%. As the whole synthesis of this arene relies on crystallization and all reagents are cheap, it is highly scalable and multigram quantities can easily be prepared (here: > 40 g of the benzyl chloride were prepared in a single run). The above mentioned alkyne was carboaluminated and after many optimization attempts the resulting alane was coupled with the benzyl chloride in a Negishi fashion to furnish the desired farnesylarene in high yield (83%). After various efforts this compound could selectively be dihydroxylated utilizing the Noe-Lin ligand. Then the obtained glycol was converted to the corresponding oxirane, subjected to a bioinspired, Ti-(III)-mediated domino cyclization and debenzylated to furnish the desired 3β-hydroxyalbicanylresorcine derivate. Testing numerous conditions and reagents phenyliodine bis(trifluoroacetate) in a cold DMF/H2O mixture shows up as the optimal system for the high yielding oxidation (91%) of this resorcine to the corresponding 2-hydroxyquinone. Subsequent desilylation revealed the synthetic target 3β-hydroxytauranin. Selective oxidation of the secondary alcohol function of the still silylated 3β-hydroxytauranin derivate besides the extreme oxidation labile 2-hydroxyquinone moiety employing Dess-Martin periodinane under smooth conditions disclosed 3-oxotauranin after desilylation. All spectroscopic data matched very well with those published for both natural compounds. Consequently, both meroterpenes could be prepared for the first time and furthermore it was shown, that the natural product is indeed 3β-hydroxytauranin and not the originally proposed 3α-hydroxytauranin.
